Explain the procedures for collecting and handling semen specimens.

Sperm collection:

• Proper collection of semen fractions is most important for perfect analysis of male fertility evaluation.

• The first portion of the ejaculation is most important, because it comprises most of the sperm.

• When the first portion of ejaculation is not collected, the volume of semen is decreased along with sperm count.

• When the last portion of ejaculation is not collected, the volume of semen is decreased along with falsely increased sperm count and the sperm specimen will not clot properly.

• Sperm specimens must be collected during the period of sexual abstinence of more than two days and not less than seven days.

• The laboratory personnel should give sterile glass as a container to collect specimen from patients.

• Specimen is reserved at 37 degree Celsius before analysis.

• The specimens should be collected by the process of masturbation or using polyurethane condoms.

• Laboratory personnel should collect specimen and record the details along with the details of semen sample of the patient and then start the liquefaction of semen as soon as possible.
