Nuclei stain:

The nuclei structure in urine sediment is enhanced with toluidine blue (0.5%) which differentiates renal tubular epithelial cells and WBCs (White Blood Cells).

• Acetic acid (2%) enhances white blood cells nuclei and lysed RBCs (Red Blood Cells).

• It differentiates red blood cells from white blood cell, oil droplets, yeast and crystals.

Therefore, both “acetic acid and toluidine blue” enhances nuclei detail. Hence, the correct answer is option.
