Reagent strip reaction of blood:
Reagent strip reaction for blood is performed to detect blood in urine.
• The principle is “heme peroxidase” activity.
Procedure:
• The reagent strip has peroxide and tetramethylbenzidine in testing area for blood.
• Hemoglobin and myoglobin’s heme component reacts with tetramethylbenzidine (chromogen) to give oxidized chromogen in the color of green-blue.
Myoglobin:
Oxygen and iron binding protein in muscle tissue.
• Color range from negative yellow ->green->positive green->blue are formed in the occurrence of free hemoglobin or myoglobin in urine.
• In contrast, when red blood cells are destroyed and contacts the pad, released hemoglobin forms a reaction which gives speckled configuration on pad.
• This speckled pattern indicates the presence of hematuria.
Hematuria:
• Existence of red blood cells (blood) in urine caused by glomerular diseases ,renal calculi, tumors, toxic chemical exposure, anti-coagulant therapy and trauma.
